Cytogenetic Analysis: modal number = 52; range = 46 to 54; This is a hyperdiploid cell line with 24 to 26 marker chromosomes common to all cells., Nine markers （M1 - M9） are common to L-M（TK-） （see ATCC CCL-1.3） and 16 to 17 are found only in A9. The latter markers include Rb（3.5）, Rb（4.15）, Rb（5.5）, T（8;19）, Rb（9.17） and 11 to 12 others., Structurally normal chromosomes 4, 15, 16, 17, 18 and X were absent.

Comments: The A9 cell line was isolated by J.W. Littlefield from wild-type Strain L cells.
The cells are sensitive to HAT selection medium and deficient in adenosine phosphoribosyl transferase （APRT） and hypoxanthine phosphoribosyl transferase （HPRT）.
The cells are resistant to 8-azaguanine （0.003 mg/ml） and 6-thioguanine （0.1 mM）.
Propagation: ATCC complete growth medium: Dulbecco's modified Eagle's medium with 4.5g/L glucose, 90%; fetal bovine serum, 10%
Subculturing: Subc*tion Ratio: A subc*tion ratio of 1:4 to 1:10 is recommended
Medium Renewal: Every 2 to 3 days
Remove old medium, add fresh 0.25% trypsin, EDTA （0.03%） solution, rinse and remove. Allow cells to sit at room temperature （or at 37C） until cells detach （about 5 to 10 minutes）. Add fresh medium, aspirate and dispense into new flasks.
